Post
translational modification of proteins (PTMs) act as key regulators of protein
function and thereby greatly contribute to the functional
diversity of the proteome. However, the analysis of PTMs is difficult due to
the heterogeneity of modification and lack of mass sensitivity. Compared to global proteomics, information
obtained through PTM proteomics is often limited. To overcome this issue, we have developed that
integrated PTM proteomics pipeline. First, we extract crucial pathways from
global proteome anlaysis, then collect PTM information on proteins present in
each pathway. Our approach allows us which PTMs to focus on in an unbiased
manner and generate vizualizations to reveal PTM sites within proteins. We investigate PTM associated relationships and
their role in pathways obtained through global proteomics in colong cancer studies.
Global proteomics revealed changes of proteins involved in ECM related pathways
targeting glycosylation as PTMs to focus on. We found that protein
glycosylation patterns were changed in colon cancer.
