The ministry of food and drug safety (MFDS) has
set its pesticides residue level limits in livestock at 0.01 mg/kg and less
through 2024, named as positive list system. Thus, it becomes necessary to
establish a robust analytical method to determine the maximum residue limit of
pesticides for these animals. Herein, we analyzed triazole fungicides, a known
ergosterol biosynthesis inhibitor pesticides, commonly used in livestock feeds.
We have developed a bioanalytical method to measure trace levels of triazole
fungicides, including flutriafol, tebuconazole, and propiconazole in daily
goats. Rats were used to construct a predictive toxicity model targeting
translocation of pesticide residues in rat muscle, fat, and plasma. We applied the
QuEChERS method to extract triazoles in daily goat and rat samples and then
measured three triazole levels using liquid chromatography coupled to tandem
mass spectrometry (LC-MS/MS). Our method provides reliable linearity (r2>0.99),
accuracy (≤15% RE), and precision (≤15% CV). Recovery tests (≤15% CV) also
demonstrate efficient extraction. Thus, our method has shown success at
determining triazole fungicide residues in biological samples that is
sufficient to meet MFDS standards.
