Rutin (quercetin-3-o-rutinoside) and quercetin are
the most common flavonols regarded as an essential components in various fields
in point of medical, pharmaceutical, and nutraceutical applications. In
addition, it is actively studied that quercetin have advantages over rutin,
such as more efficient chemoprevention, potent inhibition of nitric oxide (NO)
production, and better cytoprotective abilities. Due to theses excellence of
quercetin, the hydrolysis of rutin to quercetin by chemical reactions or
biological metabolism in microorganisms and its qualitative & quantitative
analysis, is still the active realm of research. In this work, the
HPLC-ESI-MS/MS method has been used for the flavonol specific quantitation
using naringin, which is one of flavonol family, as an internal standard.
Flavonol mixtures either prepared as standards or obtained from metabolic
pathway were quantified using the selective ion monitoring (SIM) mode in a
quadrupole ion trap mass spectrometer. The performances of quantitation, such
as the linear dynamic range, the limit of detection (LOD), and the limit of
quantification (LOQ), were defined by standard flavonol samples with a
concentration range from 1 μM to 1 mM. Furthermore, the bacterial turnover
efficiency for the conversion of rutins into quercetins could be successfully
quantified.
