Analysis of amphenicols and metabolite in milk and egg using ultrahigh-performance liquid chromatography coupled to tandem mass spectrometry

Amphenicols are a group of chemical compounds in cluding chloramphenicol (CAP), thiamphenicol (TAP), florfenicol (FF), and florfenicol amine (FFA). In this work, a modified QuEChERS extraction method followed by UPLC-MS/MS was developed for the simultaneous quantification of residues, including CAP, TAP, FF, and FFA in egg and milk. The target analytes were extracted with a combination of acidified acetonitrile and ammonium hydorxide subsequently purified with original QuEChERS kits (composed of magnesium sulfate and sodium chloride) as well as n-hexane. All analytes were separated on a reversed-phase analytical column using and a mobile phase of (A) distilled water and (B) acetonitrile with 0.1% acetic acid. Good linearity (R2≥0.9958) was achieved over the tested concentration range (0.5–5 μg/kg for CAP, 5–50 μg/kg for TAP, FF, and FFA) in matrix-matched standard calibrations. Recoveries of 69.8~102.8%, with relative standard deviations (RSDs) of ≤8.87% (intraday and interday), were obtained for each sample at three spiking concentrations (CAP:0.5, 1, 5 μg/kg; TAP, FF, and FFA:5, 10, and 20 μg/kg). In conclusion, the current approach is simple, efficient and reliable and can successfully determine the residual levels of CAP, TAP, FF, and FFA in milk and egg.
