This study was aimed to determine the changes of feces metabolites after LBP administration in healthy male Korean (n=20). Metabolites were evaluated before and after administration, according to dosage (1 x 1011, 3 x 1011, 6 x 1011, 9 x 1011 colony forming unit), and dietary control. Fecal samples were collected before and up to 6 days after administration of LBP. Untargeted metabolomic analysis was performed using ultra-high pressure liquid chromatography (UHPLC)-quadrupole/orbitrap mass spectrometer with heated electrospray ionization (HESI) source after sample preparation. Kinetex C18 column 2.1 x 100 mm (2.6 μm) (Phenomenex, Torrance, CA, USA) was used for chromatographic separation with temperature maintained at 40℃. Scan range was 100-1,000 m/z with full MS-data dependent MS2 (ddMS2) scan mode at resolution of 70,000-17,500.
A total of 22 metabolites were selected as candidates, and pathway analysis was performed using MetaboAnlyst 5.0. The analysis proved that the tyrosine, tryptophan, and phenylalanine biosynthesis pathway was most relevant (Impact value 1.0, p<0.05). The metabolites involved in this pathway, tyrosine, tryptophan, and phenylalanine, increased approximately twice each from 5-6 days after LBP administration compared to before LBP administration (p<0.05). This study results provides metabolic changes after LBP administration in healthy subjects.
