Artemisia capillaris is widely used for its anti-inflammatory and hepatoprotective properties. However, its complex matrix and numerous structural isomers hinder precise quantification. achieving complete chromatographic resolution via HPLC is a crucial prerequisite for accurate MS detection. This study developed and validated a rapid, accurate high-performance liquid chromatography method to simultaneously determine six marker compounds: Neochlorogenic acid, Chlorogenic acid, Cryptochlorogenic acid, Caffeic acid, 3,4-Dicaffeoylquinic acid, and Scoparone. Analysis utilized an optimized gradient elution on a reversed-phase column with mobile phases . The method was rigorously validated for specificity, linearity, precision, and accuracy, achieving perfect baseline separation of the target isomers. Finally, it successfully evaluated the phytochemical profiles of four A. capillaris samples. By completely resolving these isomers, the optimized HPLC conditions provide a robust foundation readily transferable to LC-MS platforms, enabling ultra-trace quantification and advanced bioanalysis in complex biological matrices. |
