Wild pear (Pyrus spp.) is a valuable genetic resource with nutritional and functional potential; however, metabolomic information on different fruit tissues remains limited. This study compared the metabolite profiles of peel and flesh from three wild pear samples using untargeted UPLC-QTOF-MS in positive and negative electrospray ionization modes. Freeze-dried tissues were extracted with methanol/acetonitrile/water containing an internal standard and analyzed using an Agilent SB-AQ column coupled to a SCIEX X500R quadrupole time-of-flight mass spectrometer. Quality control (QC) samples and procedural blanks were included throughout the analytical sequence. Metabolites were putatively annotated using the SCIEX OS spectral library and classified into major metabolite classes. Positive ion mode yielded more annotated metabolites than negative ion mode, and the peel consistently contained more annotated metabolites than the flesh. The annotated metabolites were mainly classified as organic acids (n = 62), lipids (n = 54), flavonoids (n = 36), terpenoids/triterpenoids (n = 35), phenolic acids (n = 32), carbohydrates (n = 30), and amino acid derivatives (n = 19). Flavonoids, phenolic acids, and terpenoid-related metabolites showed greater diversity in the peel than in the flesh, indicating distinct tissue-specific metabolite compositions. These findings provide fundamental metabolomic information on wild pear fruit tissues and support future studies on their nutritional and functional utilization. |
