Pantothenic acid (vitamin B5) is present in a wide range of foods, which serve as the principal dietary source of this vitamin. Its accurate quantification in foods is vital for the provision of reliable nutritional information. Although a variety of methods have been reported for pantothenic acid analysis, most have been validated with limited food matrices. Given the compositional complexity of food matrices, the development of accurate analytical methods applicable to a broader range of foods is required to meet nutritional demands. An isotope dilution–liquid chromatography–tandem mass spectrometry (ID-LC-MS/MS) method using [13C3, 15N]–calcium pantothenate as an internal standard was developed for the accurate determination of pantothenic acid in meats and grains. This method employed 10 mM ammonium acetate (pH 5) extraction combined with protein precipitation, thus eliminating matrix interference. When validated with beef, rice flour, and wheat flour, the method achieved an accuracy of 99.7–100.8% with intra- and inter-day relative standard deviations < 1%, and measurement uncertainties generally < 3%, demonstrating significantly improved analytical performance compared to existing methods. The developed method was applied to various food materials (beef, pork, chicken, rice, wheat, and corn flour) purchased from a local market. |
