In antibody–drug conjugates (ADCs), Fc N-glycosylation is a critical quality attribute, and comparing glycan profiles across products offers useful insight for characterization. We established an LC–MS workflow to identify and compare the Fc N-glycans of trastuzumab (Herceptin) and four ADC standards (Kadcyla, Enhertu, Trodelvy, Adcetris). Released N-glycan profiling (RapiFluor-MS, HILIC-UPLC-FLR) assigned major glycans by retention time, and TCEP-reduced heavy-chain intact mass gave orthogonal confirmation through the characteristic +162 Da (galactose) and −146 Da (fucose) spacing. Both methods identified a G0F-dominant, fucosylated biantennary set (G0/G0F/G1F/G2F). Herceptin and Kadcyla were nearly identical, whereas Enhertu—despite the shared trastuzumab backbone—was markedly enriched in G0F, and Trodelvy and Adcetris showed product-specific distributions. Pairing released-glycan and intact-mass analysis provides a robust orthogonal approach for Fc N-glycan identification in ADC standards. |
