Multiple myeloma is characterized by the abnormal proliferation of clonal plasma cells and a corresponding increase in monoclonal immunoglobulin, referred to as M-protein. Aberrant glycosylation of M-protein light chains may be clinically relevant because it may reflect clonal biology and disease progression. Current routine clinical tests, including protein electrophoresis (PEP) and immunofixation electrophoresis (IFE), can identify M-proteins but cannot directly assess their glycosylation. In this study, we detected glycosylated light chains in M-spike-positive samples through MALDI-TOF and Q-TOF analysis. Furthermore, serial samples collected 61 days apart from the same patient showed an identical glycosylated light-chain feature, supporting its potential as a clone-specific marker. The persistence of this glycosylation pattern suggests that it may represent a molecular feature of the disease-driving plasma-cell clone in multiple myeloma patients. |
