Steroid hormones play pivotal physiological roles in the endocrine system and profiling analysis of steroid hormones in various biological specimens is crucial for elucidating their metabolic alteration. This study evaluated and compared the analytical performances of GC-MS and LC-MS for urinary steroid profiling. For sample preparation, 400 µL of urine was purified using solid-phase extraction, hydrolyzed with β-glucuronidase, and extracted by liquid-liquid extraction. The resulting residues were analyzed by GC-MS following chromatographic separation on an MXT-1 capillary column (30 m × 0.25 mm ID, 0.25 μm i.d.) with detection in MRM mode as their trimethylsilyl derivatives. For LC-MS analysis, the resulting residues were reconstituted in methanol and centrifuged on an ultra-free MC centrifugal filter. The filtrate was then separated on a Hypersil GOLD C18 column (50 mm × 2.1 mm, 1.9 μm) and analyzed in ESI/MRM mode. A total of 32 steroids (14 androgens, 9 corticoids, and 9 progestins) and 31 steroids (8 androgens, 19 corticoids, and 4 progestins) were detected by GC-MS and LC-MS, respectively, while 7 estrogens were detected exclusively by GC-MS. Overall, GC-MS demonstrated superior detectability for androgens, progestins, and estrogens, whereas LC-MS showed better performance in corticoid analysis. |
