Food contact materials(FCMs) are materials that contact food during handling and storage. Among FCMs, intentionally added substances(IAS) are strictly regulated, but non-intentionally added substances(NIAS) can form through secondary reactions or elution, leading to poor control and limited information. NIAS migrating into food may pose ingestion risks. In this study, the FCMs extraction method was optimized and a non-targeted analysis(NTA) of NIAS was performed using liquid chromatography-high resolution mass spectrometry(LC-HRMS), which is specified by high sensitivity and accuracy. The FCMs extraction method was optimized by comparing the number of NIAS compounds assigned to each extraction factor(extraction solvent and time) using LC-HRMS non-targeted analysis data, with the highest NIAS compounds detected when methanol was extracted for 45 minutes. Using an optimized extraction method and LC-HRMS, NTA of FCMs on 150 food utensils, containers, and packages were performed to establish a NIAS database for each sample. To select new hazardous candidates in FCMs, multi-frequency analysis and oral/dermal toxicity in-silico assays were performed based on the NIAS database. New hazardous substances causing oral/dermal toxicity, including phthalates, fatty acids, and phenyl compounds, were tentatively identified in the NIAS, suggesting the need for monitoring to improve the safety management of FCMs.
