Plant hormones, or phytohormones, are key regulators of plant growth, development, and environmental responses. Accurate and rapid quantification of these signaling molecules in complex plant matrices requires efficient sample preparation and sensitive analytical techniques. In this study, a modified QuEChERS (Quick, Easy, Cheap, Effective, Rugged, and Safe) method was combined with ultra-performance liquid chromatography coupled with a QTRAP mass spectrometer (UPLC-QTRAP MS) to simultaneously quantify multiple plant hormones. Extraction was performed using 80% acetonitrile with 0.1% formic acid, followed by a dispersive solid-phase extraction (d-SPE) clean-up to minimize matrix effects. The method enabled simultaneous detection of auxin (IAA), cytokinins, gibberellins (GA1, GA3, GA4, GA7), abscisic acid, salicylic acid, and jasmonic acid in a single run. Analysis was conducted using electrospray ionization (ESI) in negative ion mode, with chromatographic separation on a C18 reversed-phase column under optimized gradient elution. The method demonstrated high sensitivity, good linearity, and reproducibility, with detection and quantification limits suitable for trace-level analysis. This QuEChERS–LC-MS approach is ideal for high-throughput profiling of phytohormones and applicable to studies in plant physiology, agriculture, and environmental science. |
