Plant-derived peptones are emerging as promising functional biomaterials due to their rich content of bioactive peptides. In this study, we developed a standardized and optimized process for peptones from soybean, wheat, and rice, focusing on the impact of processing conditions on the resulting peptide profiles. To achieve precise quantification and
characterization of functional peptides, we established a targeted LC-MS/MS
method using triple quadrupole mass spectrometry (QQQ). A total of 18 low-molecular-weight peptides
with known biological functions were selected and quantified under multiple
reaction monitoring (MRM) mode. Method reproducibility was validated by
evaluating intra- and inter-batch coefficients of variation (CVs), ensuring
high analytical reliability. Our results revealed significant variation in peptide profiles depending on plant type and processing conditions, underscoring the importance of process optimization for improving the yield of functional peptides. The platform demonstrated here provides a robust analytical and preparative approach for peptone production and can be extended to other plant-based protein sources. The study highlights the utility of QQQ-based peptide profiling in biomaterial development and functional food research. |
