Glucosinolates (GSLs) are health-promoting secondary metabolites predominantly found in broccoli. In this study, a total of 12 GSLs were identified using ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-QTOFMS), and three major GSLs-glucoraphanin (GRA), glucobrassicin (GBS), and neoglucobrassicin (NGBS)-were selected for quantitative analysis. To enhance their accumulation, we utilized a biotechnological tool based on regulatory insights into GSL biosynthesis, which resulted in the promotion of both aliphatic and indole GSL pathways by upregulating related biosynthetic genes. Additionally, molecular networking analysis coupled with RDBE-based structure prediction facilitated the identification of two new GSLs. Thus, the enhanced accumulation of main bioactive GSLs and the identification of new potential candidate GSLs in broccoli sprouts are anticipated to support nutraceutical research. |
