Prostate-specific antigen (PSA) is a glycoprotein widely used in clinical practice for prostate cancer (PCa) screening. However, serum PSA levels can also be elevated in benign prostate conditions, which can lead to a high false-positive rate in PCa screening. PSA contains a single <i style="font-family: "Times New Roman", serif; font-size: 12pt; text-indent: 12pt;">N</i>-linked glycosylation site, and its <i style="font-family: "Times New Roman", serif; font-size: 12pt; text-indent: 12pt;">N</i>-glycosylation is known to undergo aberrant alterations in the cancer group compared to the non-cancer group. However, blood generally contains trace amounts of PSA, making it difficult to identify the protein itself including its post-translational modifications. Therefore, this study aimed to establish sample preparation and analytical methods for the effective identification of PCa-specific PSA using a PSA standard. The PSA standard will be processed using four different preparation protocols that differ in PNGase F treatment strategies and denaturation conditions. After tryptic digestion, samples will be analyzed by nLC-MS/MS and then proteins will be identified using a protein database search software. |
