Short-chain fatty acids (SCFAs), bile acids, tryptophan, and other metabolites associated with gut microbes are known to influence immune cell regulation and the onset of various diseases. Research in this area is underway. However, analyzing metabolites derived from human intestinal microorganisms and researching how they are influenced by various factors, such as diet and lifestyle, remains an unresolved issue. In this study, CA and LCA, bile acids linked to diseases like asthma, urticaria, and sepsis, were chosen as target metabolites. The study also established methods for collecting and pretreating healthy human fecal samples and measuring and analyzing them using LC-TQ-MS through experiments. The results of the experiment confirmed the optimal collection
method and storage temperature for fecal samples, which depended on the type of
metabolite. The experiment also established the appropriate extraction solvent,
homogenization method, and precipitation time for pretreatment. In this
analysis, the LOD(ng/ml) for CA and LCA was 0.498 (SD= ±0.10009) and 2.25 (SD=
±0.10995), respectively; the LOQ(ng/ml) was 1.51 (SD = ±0.3033) and 6.81 (SD=
±0.3331) and the ME(%) was 100 ± 0.0338, 109 ± 0.0223 respectively. This study aims to develop a quantitative measurement and analysis method for human fecal metabolites based on targeted LC/MS/MS and to develop a fecal metabolite measurement method that can be applied in actual diagnostic medicine for diseases based on this method. |
