Chitin is a water-insoluble cationic amino polysaccharide of β-1,4-linked N-Acetyl-D-glucosamine (GlcNAc) whereas chitooligosaccharides (COS) are derivatives of chitin composed of monomers and oligomers of deacetylated and N-acetylated glucosamine. COS with a low degree of polymerization (DP) are water-soluble and have been reported to possess a wide range of bioactivities. However, these investigations have been often conducted with oligomer mixtures that are relatively poorly defined, thus making it difficult to distinguish COS molecules responsible for the biological effect. Thus, high-resolution liquid chromatography (LC) techniques can be utilized for the separation of complex mixtures and hyphenated with tandem mass spectrometry (LC-MS/MS) for comprehensive characterization of the molecular weight, degree of acetylation and fragmentation patterns of the separated oligosaccharide compounds. The first part of the study involved optimization of parameters for the production of COS by microbial degradation of colloidal chitin using novelStreptomyces chilikensis RC1830 isolated from the brackish water sediments of Chilika Lake, India. The crude COS extract was further investigated for in vitro anti-radical and anti-inflammatory properties. The last part involves the separation of the crude extract and comprehensive characterization using LC-MS/MS analysis. The low molecular weight COS obtained was successfully separated and the degree of acetylation of the COS was determined. The characteristic fragment patterns of COS were also obtained from the MS/MS analysis.
