The efficient extraction of lipid species in biofluids was developed for quick sample preparation and accurate results of lipid analysis, using spin column with superabsorbent polymer beads (SAP). The modified (mSAP) method is composed of simple steps, including sample loading and swelling, solvent addition, centrifugation and taking lipid extracts. The mSAP method was performed about 20 times more rapid than a conventional methods. The limit of detection (LOD) of 1,2-diheptadecanoyl-sn-glycero-3-phophocholine (PC 17:0/17:0) spiking in plasma by the mSAP method is about eight times lower than that of the conventional methods. The relative standard deviation value of inter- and intra-day of the mSAP method was higher about 3 times than the previous SAP and about 2 times than modified Folch methods over a 5-day period. Among the seven species in the lipid standard mixture, our developed method had a higher recovery rate. Based on these results, the mSAP method is more time-saving, sensitive, reproducibleility, reliable, and comfortable than modified Folch and the previous SAP methods . This method showed great potential as tools for the application of total lipids analysis for rapid disease diagnosis and lipidomic research in future.
