The
identification of N-glycosylation sites is dependent on the number of possible
glycosylation sites. Most antibodies have one or two of N-glycosylation sites,
thus it is easily identified with trypsin and LC-MS/MS analysis. However, it is
difficult to analyze the glycosylation sites of proteins containing more than 5
N-glycosylation sites with trypsin and LC-MS/MS because the glycopeptides are
too big to be analyzed. To overcome the issue, we used protease K for digestion
because the protease K produced smaller peptides and generated glycopeptides
with appropriate size for the analysis. As results, one of protein containing 6
possible glycosylation sites was analyzed and identified five glycosylation
sites.
