As a protein biomarker, prostate-specific antigen (PSA) has been widely used for the early diagnosis of prostate cancer (PCa) in clinics. However, PSA also increases in other prostate diseases, such as benign prostatic hyperplasia (BPH) and prostatitis. This leads to a higher number of false positives and causes unnecessary biopsies, an invasive procedure which is associated with risks including bleeding and sepsis. This study intended to compare PSA ranks in tissue proteomes from prostate cancer patients with PSA values in their blood to discover relationship between blood test in clinics and cancer tissues for a more accurate early diagnosis.
PCa tissues were collected from the twenty PCa patients with different PSA levels in blood test. All PCa tissues were homogenized, and proteins were extracted from them without detergent. Then the supernatant fluid was trypsin digested and desalted respectively, and the proteins in all PCa tissues were identified and quantified with nLC-MS/MS and protein database search software. For analysis, PSA ranks in cancer tissues and PSA values in blood were compared according to the Gleason grade which evaluates the malignancy of prostate cancer.
